This publication is designed to explore how digital text is different from old and familiar printed or handwritten text. There is a vast amount of information available in our digital lives, found through the internet, filed under numerous categories and of varying quality, all within the worlds sight. All this digital information is disjointed from the traditional linear form of progression. When adapting stories found on the internet for this book many design aspects were taken into consideration. One of the most visible is the lack of a table of contents. Instead of a table of contents the book instructs the user to remove the binding clips and open the book into poster form and step back and view the journey as a whole. The publication was inspired by and incorporates the ideas set forth by Michael Wesch, Professor at Kansas State University. Wesch asks many questions in regards to who creates the digital content and what rights they and others have to this content. I agree these questions need to be considered when determining the value of this ever changing and growing form of digital information and collaboration, which varies according to the needs of the user. This publication examines these ideas by taking several stories out of the non-physical realm and publishing them in the physical material form of a book. The stories vary in length, quality, and importance, thereby communicating how critical it is to remember digital text is different in that anyone can publish digital text. We must continuously ask how to handle the integrity of digital text for future generations and are only able to dream where future generations take it next.

EXAMPLES of specific Elaboration questions from MATH You're studying calculus. The topic is “derivatives”. How do derivatives work? Well, they are the rate of the change. How does that work? You take a look at one point, then you take a look at a prior point, over some interval. And then you take the difference divided by the interval. As that interval approaches zero, you have the instantaneous rate of change. Why does this happen? Because “instantaneous” means that the interval is nothing. SCIENCE Imagine you are studying neural communication, maybe in a biology, neuroscience, or psychology class. How does neural communication work? If we look at one neuron, the dendrites receive messages from many other neurons, and then the messages converge in the soma. If there is enough of a positive charge within the soma, then an action potential will occur, and an electrical signal goes down the axon. When the signal reaches the terminal buttons, neurotransmitters release into the synapse where they communicate with the dendrites of the next neuron. Why does this happen? The neurotransmitters are chemicals that allow neurons to communicate with one another. The pattern of activation among different neurons (which neurons fire, how quickly, what neurotransmitters they release) determines the message in your brain. You might then ask, how does the axon work? The axon is a long tail-like structure that produces the electrical signal. How does the signal travel? The axon is covered in myelin sheath, a fatty substance that insulates the axon. The myelin sheath works like the rubber around the cord of an electrical appliance, and it serves to make the electricity travel faster. Why have myelin sheath? Because we need our neurons to be able to send signals fast, since we need to be able to react, make decisions, move quickly, perceive feeling in our skin instantly, etc. Make sure to compare ideas to learn how they are similar and different. For example, an axon and terminal buttons are both parts of a neuron; but, the axon sends an electrical signal while the terminal buttons release chemicals. Both Schizophrenia and Parkinson’s disease are related to the neurotransmitter dopamine, but Schizophrenia is the result of too much dopamine while Parkinson’s disease is the result of too little dopamine. Also, try to make connections to your own memories or experiences, and compare ideas to learn how they are similar and different. We already made the connection from myelin sheath on axons to the rubber on cords to electrical appliances. Here is another example: a family member or close friend who suffers from Schizophrenia disease is suffering from too much dopamine. This means that too much dopamine is being released, by the terminal buttons, into the synapse. A doctor could give them a drug to reduce the dopamine in their brain, called a dopamine antagonist. If too much of this drug is used, the patient might begin developing symptoms similar to Parkinson’s disease. How would a dopamine antagonist work? … continue asking yourself elaborative questions! HISTORY Imagine you’re studying World War II, and the attack on Pearl Harbor. You could ask yourself, how did this attack happen? On December 7, 1941, the Imperial Japanese Navy attacked the United States Naval Base at Pearl Harbor. The attack included Japanese fighter planes, bombers, and torpedo planes. Why did this happen? The Japanese intended to destroy the United States’ Pacific Fleet so that it could not interfere with Japanese operations. Here you could also ask another type of question: What was the result of this historic event? Well, Japanese casualties were light, while they damaged eight U.S. Navy battleships. The Arizona was among those that the Japanese sunk, and was not raised from the shallow water. U.S. aircrafts were also destroyed, and 2,403 Americans were killed (1,178 were injured). Why is this event important? The day after the attack, Roosevelt delivered his Infamy Speech, the United States declared war on Japan, and Japanese-Americans were then relocated to internment camps. You could then go on: how did the U.S. enter the war? How did the Pearl Harbor attack lead up to the release of the atomic bomb? How did the war end? And so on. There are so many ways to explain the idea and add details!

How to Make a Movie Poster - Design Theory --------------------------------------------------------------------- A movie poster is a filmmaker's first line of attack when getting people to know about your film. Unlike a movie trailer, you can put your poster in as many places as you want. The idea behind a movie poster is to get people to want to see your film. You should be thinking about that fact throughout your design ideas. As you begin to sketch out or visualize your design, you want to think about the genre of your film. This seems like a no brainer, but it's really important. Everything you do with the poster will depend on the genre. Next, you need to determine the mood of the poster within its genre. For example, a horror movie could be a slasher horror or a thriller horror. A slasher contains a lot of violence, but a thriller tends to contain less violence. You may or may not want to depict the mood of the film by portraying that. You then want to decide which of the cast members should be in the poster. This can range from no cast members to a full ensemble. It all depends on the movie, but generally it is a good idea to show who is starring in your movie, especially if it is someone an audience will recognize.  Let's analyze a famous poster like one from the Star Wars series to get an idea of how to put one together. The Star Wars posters are staples of traditional classic movie posters that feature the lead cast along with a few other elements that are important to the movie. First, the background is black space. This is done to make the more colorful elements stand out and to imply that the movie takes place in space. Second, there is a large use of bright light. As you can see, the two suns of Tatooine shine brightly between all of the characters. The shine and flare of the suns creates a sense of fantasy, imagination and grandeur. Put simply, it makes it look like a really big deal. Third, the characters spread throughout the poster are shown in different ways to convey their importance to the story. Anakin Skywalker is the little boy in the center. He is the focus of the film. The eyes will always focus on the middle of the poster, so important elements should be centered. Above and to the poster's left is a big head shot of Qui-Gon Jinn, played by Liam Neeson. There are a few reasons he was placed here: he is the mentor to both Anakin and Obi-Wan Kenobi, he led on the plot more than anyone in the movie and he served as the emotional core of the story. Either those, or he was simply the top billed of the cast, meaning he was paid the most. This is unlikely, but there are plenty of situations where the top billed actor gets featured on the poster more than anyone else. Usually, it's the main character, so it works out fine. To the poster's right is Queen Amidala. Her unique appearance already creates interest. Where is she from? What is her significance to the story? She is the only woman on the poster, so it can be assumed that she might be the romantic interest. To the lower right are Obi-Wan Kenobi and Jar Jar Binks. Obi-Wan is shown holding his lightsaber. This is done to let the audience know two things: the kind of character Obi-Wan is in the film, and the lightsaber itself reminds people that those awesome lightsabers will be seen in the movie. Even the way Obi-Wan holds the lightsaber denotes a certain mood. Jar Jar is smaller, indicating a more supportive role, but his odd look and goofy smile implicate that he is the humorous character. To Anakin's left are the Naboo's ship (Queen Amidala's transport), R2-D2 and C-3PO. Notice that C-3PO has no visible plating. This adds to the sense of wonder and makes you want to see the movie. The ship next to the droids accentuates the science fantasy nature of the film. It looks different than anything any of the previous Star Wars movies have seen. The ship is sleek, beautiful and streamlined. Very little in the previous films had designs like this, implying that we will see a new vision of Star Wars with new ships, environments, and other things that the audience will look forward to seeing. Finally, a bright red and black face with menacing orange eyes covers nearly the entire background of the poster. This is the villain of the movie: Darth Maul. The crimson red colors and the... Because I know what I like when I look at it, but I have no idea how to come up with what would work. So, you may be someone who has a lot of design ability, and can at least begin the idea. Or, you may be like me, and need to hire a designer to help you create exactly the right menu for your restaurant, for your concept. Some things to consider: size. You don't want the menu to be so big, and so designed, that it's a pain in the neck. You don't want it to become the additional guest at the table. You don't want it to be something that blocks my ability to see my friends across the table. And, on the other hand, you don't want it to be so small that people who are of a certain age can't read the type. So, it has to be the right size. It has to be able to hold all the information you want it to hold in a legible and comfortable way. Then, there are a lot of branding decisions. Will you have your logo on this menu? What colors will be used on the menu? What kind of physical format will it have? Will it be paper? A certain texture of paper, or color of paper? Will it be in a very deluxe leather folder? What will the physical menu feel like to hold, and what impression will it create as the guest uses it? You also need to think about layout of the menu. Will there be appetizers, entrees, snacks, sides, salads, pastas? What sections of information will there be? Will you want to put cocktails on the food menu? Will you want to put desserts on the food menu, or have a separate menu for dessert? So, what sections does your menu need to have? And also, remembering that your menu is a very powerful tool for marketing your restaurant. What information should be on it? Should the address, phone number, website be on the menu, so that when someone prints it online, they have all that information with them? In menu design, it's important to remember that your menu is your principle sales tool. It's what the guests use to choose what they're going to buy, so where their eye goes and what their eye sees is vitally important. It's also really important to remember that you want the lyrics, or the words that are used to describe your menu items, to be appealing and just the right amount of information. You don't want so much information that their eyes glaze over as they're reading it. You don't want so little information that they have no confidence that they know what they're ordering. So, writing the menu words, which a designer probably can't help you with, needs to be something you think about a great deal. What is going to make people both want to order the item, but also give them real confidence that they're going to get what they thought they were ordering?

Develop Africa is working with CAPEC, a local non profit / NGO organization, helping to provide books, teaching supplies, school supplies and scholarships. Books are one of the foundational tools for learning. Many of these children did not have any books at home. The school at that time did not have an adequate range of books that they could use to promote reading. Through these books, the children are developing a love for reading and learning -- which are important keys to success in life. The primary language of children in this area of Cameroon is French and by enhancing their knowledge of English, the children are being better prepared for life as global citizens.

With handwriting like that, would you believe me? People come to your website to read your content. Blog posts, your opinions, explanations. People visit your website to learn information from. Newsflash! People don’t come to your website just to you have cool colors or some nice designs….unless you’re a photographer or some really cool artist. They come for your content. And that's why good typography is important. If your content is tough to read, people will have difficulties reading or not take you as serious. And we don’t want that. Typography influences your web visitors and can have positive effects on your business when done correctly. Some feel cheerful. Some feel energizing. Some feel creepy. An experiment was conducted with 40,000 people. The peeps were split into groups and presented the same passage but with different typefaces. The readers were then asked whether they agreed to disagreed with the passage. The findings, people reading the statement in one font we're much more likely to agree with the passage than others reading the same thin in another font. Study below in the description. Conclusion, typography affects readers. But don't take the studies word for it. Ask your friends see what they say about fonts that you're using on your website. Because if you’re typography is hard to read or not matching your style, it might be off-putting. It might be distracting enough to not talk to you. What is accuracy in Next-Generation Sequencing (NGS)? – Seq It Out #2

Ask your question at https://www.thermofisher.com/ask Whether you are new to next generation sequencing, or an old hand, you have likely encountered terms like base quality, Q-score, consensus accuracy and so on. This could have happened when you were reading a NGS review or listening to a bright-eyed grad student presenting her new variant calling algorithm at a poster session. Either way, you may have been left thinking, “Huh?” Let’s turn that “Huh?” to an “Oh, yeah” while we take a closer look at the statistics of sequencing So let's begin our journey from the good old days of base quality scores, also known Q-scores. Back in the early days of DNA sequencing, shotgun sequencing was typically employed to perform de novo assembly. Basically, DNA was fragmented into shorter pieces and sequenced. The resulting sequencing reads were then assembled to build a reference genome. Since we didn't know what the truth was back then, we tried to predict the quality of the sequenced bases using a, well, de novo methodology. That is, by doing some complicated signal analysis and assigning a base quality score on Phred scale, or a Q-score, to every base observed. The Q-score is a value derived from the formula Q equals negative10 log P, where P is the probability that the base is wrongly called. A Q-score of 10 translates to 1 in a 10 chance of the base call being wrong, Q20 is 1 in 100 chance And Q30 is 1 in 1000 chance. What I just described is what we refer to as the predicted quality in the sequencing world. You are likely have encountered a metric called %bases greater than or equal to Q30 which translates to the percent of sequenced bases that have a predicted quality score of 30 or more. Moving to the present, we now have the benefit of knowing the reference genome. As a result, we can take individual reads, map them to a reference and count the number of mismatches. This gives us the actual observed measure of raw read accuracy, or the empirical quality, measured by the error rate. An error rate of 1% translates to the raw read accuracy of 99% and tells us that one in a hundred bases is a mismatch to the reference. Keep in mind, that error rate will count actual biological variants as mismatches too and; thus is a conservative metric of observed quality. Determining true variants from base calling errors is where the consensus accuracy comes into play. One of the advantages of NGS is the ability to produce millions of sequencing reads. As you can see, when we pile these reads up together along the reference, what we get is the power of majority. We can filter out mismatches that are likely to have been base calling errors and keep those that are likely to be real variants using statistical inference. What we get in the end is a consensus sequence and its corresponding consensus accuracy with respect to the reference. Hopefully you are now familiar with the idea of Q-scores, raw read accuracy, and consensus accuracy as it pertains to NGS. But I am sure you’ll have more questions.
